That really depends on the requirements of the application you're going to write.
There is no best fits-them-all solution.

I have about 12 years experience developing mid-large size web applications. Here are my preferences:

1. Ruby on Rails
2. CakePHP (PHP based MVC framework)
3. Yii Framework (PHP based MVC framework)
4. Zend (I would have listed it at #1 or #2 however Im listing it at #4 due to non-existent support online)
5. JSP - Java Server pages is good if you would want your web application to work along with other ERPs active in your enterprise, since Java is platform independent.

One of my favorites is Jquery. Its a great language to learn if you want to move stuff around in the DOM without refreshing the web page.
